Spot is Plankton's pet amoeba. He first appeared in the episode Plankton's Pet, and it is his first out of four appearances on the show so far; he also appears in Plankton: The Movie. Plankton wanted him to be a guard dog but, due to Spot being nice, he decides to make him a retriever. Spot is smaller than Plankton and appears as a little speck as seen when SpongeBob was holding a picture. He also has the ability to grow in size.

- He is seen with two eyes in "Plankton's Pet" and A Place for Pets.
- This may suggest that Spot has two eyes and not one.
- Spot is the first supporting character to permanently reside at the Chum Bucket.
- Spot is the third occurrence of amoebas seen in the series. The first appearance of amoebas is in Patty Hype where Mr. Krabs watches a show about them on television, and the second appearance of amoebas is at the end of the episode Goo Goo Gas.
- After Spot's debut in "Plankton's Pet," he was absent for over four years before returning in Spot Returns.
- Spot is the only character that acts like a dog besides worms and occasionally, Patrick.
